In this article, we derive several simple approximations (confirmed by simulations) for the mutation load at high mutation rate (U), using a general model that allows us to play with the number of selected traits (n), the degree of pleiotropy of mutations, and the shape of the fitness function (which affects the average sign and magnitude of epistasis among mutations).

Epistasis occurs when one gene affects the phenotypic expression of a second gene: One gene turns on/off production of a pigment while another pigment controls the amount or color of the pigment. Pleiotropy occurs when one gene has more than 1 phenotypic expression.

Genes can either mask each other so that one is considered “dominant” or they can combine to produce a new trait. It is the conditional relationship between two genes that can determine a single phenotype of some traits. Epistasis due to recessive genes is called recessive epistasis. In mice albinism (white coat) is produced by a recessive gene aa. There is a different gene B which in the dominant state (BB and Bb) produces grey coat colour called agouti, and when recessive (bb) leads to black coat colour. Main Difference – Pleiotropy vs Polygenic Inheritance Both pleiotropy and polygenic inheritance are two terms used to describe the relationship between genes and their phenotypes or traits.

For a two locus, two allele system, there are eight independent types of gene interaction. Epistasis is the interaction between genes that influences a phenotype. Genes can either mask each other so that one is considered “dominant” or they can combine to produce a new trait. It is the conditional relationship between two genes that can determine a single phenotype of some traits.

Epistasis vs pleiotropy

Epistasis is a type of non allelic gene interaction where masking on one gene occurs by another gene while polygenic inheritance is a phenomenon where 2 or more gene together controls a phenotypic character. In another words u can say epistasis involves suppression or masking of gene by another while no such masking occur in polygenic inheritance.

Epistasis vs pleiotropy

Sign epistasis is a central evolutionary constraint, but its causal factors remain difficult to predict. Here we use the notion of parameterised optima to explain epistasis within a signalling cascade, and test these predictions in Escherichia coli.

Evolutionary adaptation is often likened to climbing a hill or peak. While this process is simple for fitness landscapes where mutations are independent, the interaction between mutations (epistasis) as well as mutations at loci that affect more than one trait (pleiotropy) are crucial in complex and realistic fitness landscapes.
